Lxvi. the Estimation of Carnosine in Muscle Extract-a

چکیده

IN a recent communication [Hunter, 1921] a colorimetric method for the estimation of carnosine in muscle extract was generally outlined. This was based on a method of estimation of iminazoles devised by Koessler and Hanke [1919, 1]. This method depended on the coupling of carnosine with diazotised sulphanilic acid in alkaline solution to form an azo dye. The colour produced was measured against a standard solution of a mixture of Congo red and methyl-orange. Pure solutions of carnosine were found to react very satisfactorily, and in different extracts of the same muscles the colour values were consistent. The accuracy of the method could not be doubted, but the certainty that carnosine was responsible for the total colour production was called in question. The problem-of the extent to which carnosine is responsible for the production of the azo colour in muscle extracts-may be attacked in two ways: 1. By the elimination of all interfering substances. 2. By a direct method of confirming the carnosine content.

Carnosine and Anserine in Mammalian Skeletal Muscle *

Two imidazole derivatives, carnosine and anserine, have been isolated from vertebrate skeletal muscle. Carnosine may be readily obtained from the ox (l), horse (2), pig (3), etc. Anserine, a methyl carnosine, has been isolated from birds, a reptile (4), fish (5), and certain mammals (6). The presence of the two compounds in muscles from the same animal has been adequately demonstrated only once...

Quantitative Studies of Carnosine and An- Serine in Mammalian Muscle* I. a Method for the Determination of Carnosine and Anserine

Carnosine was first isolated from beef muscle extract by Gulewitsch and Amiradsibi (10) in 1900, and has subsequently been shown to have a wide-spread occurrence in mammalian skeletal muscle. The investigations of Gulewitsch (8, 9), Baumann and Ingvaldsen (3), and Barger and Tutin (2) proved that carnosine is /3-alanyl-Lhistidine. Anserine was isolated from an extract of goose flesh by Ackerman...

Concerning Histidine and Carnosine

Our knowledge of carnosine is largely due to the admirable investigations of Gulewitsch. The discovery of the base in beef muscle extract was first reported by Gulewitsch and Amiradiib? in 1900. Later Kutsche? isolated a substance from beef muscle extract, which he named ignotine, but Gulewitscl? showed that ignotine and carnosine were identical. In the same volume, he4 showed that hydrolysis o...

Heat and ultrafiltration extraction of broiler meat carnosine and its antioxidant activity.

This study examined the effects of extraction and further ultrafiltration on the carnosine content, antioxidant activity and total iron content of chicken muscle extracts. Fresh breast meat had 7-fold higher carnosine than fresh thigh meat (2900 versus 419μg/g meat, respectively). Carnosine extracts of breast and thigh were prepared by heating at 60, 80 and 100°C, and ultrafiltration (UF) using...

The participation of pyridoxal phosphate, carnosine and calcium in the reversal of contraction in muscle fiber systems.

A relaxing system requiring phosphocreatine as the relaxant may be restored to glycerinated psoas fibers by incubation in a reaction mixture containing pyridoxal phosphate, calcium, carnosine, magnesium, ATP, and sodium maleate at pH 6.5. The capacity to relax, once established, persists after elution in buffer of the diffusible fraction of the reactants. The relaxation by phosphocrestine is re...
